ASTM D6319 vs. EN 455: Decoding the certification standards for Nitrile Examination Gloves.
ASTM D6319 and EN 455 are both commonly mentioned when purchasing nitrile examination gloves, but they are not two interchangeable certificates.
ASTM D6319 is a specific performance specification for nitrile examination gloves.
EN 455 is a multi-part European standards series covering medical gloves for single use, including examination and surgical gloves.
For procurement teams sourcing for both US and European markets, understanding this distinction is more useful than asking whether one standard is “stricter.”
What ASTM D6319 Covers
The current ASTM edition is ASTM D6319-19(2023).
ASTM states that the standard applies to nitrile rubber gloves used for medical examinations and diagnostic or therapeutic procedures. It covers packaged sterile, non-sterile, and bulk non-sterile nitrile examination gloves.
Requirements address areas including:
- Freedom from holes
- Dimensions
- Physical properties
- Tensile strength
- Ultimate elongation
- Performance after ageing
- Powder content
- Sterility where applicable
The FDA currently recognizes ASTM D6319-19(2023) as a consensus standard for nitrile examination gloves.
However, an ASTM test report alone is not the complete US market-entry requirement.
FDA regulates medical examination gloves as Class I reserved devices requiring 510(k) premarket notification.
EN 455 Is a Series, Not One Test
When a supplier says that a glove “meets EN 455,” procurement should ask which parts have been evaluated.
The current series includes several separate areas.
EN 455-1:2020+A2:2024 addresses freedom from holes and the related test method.
EN 455-2:2024 covers physical properties of examination/procedure and surgical gloves, including characteristics needed to maintain adequate protection during use.
EN 455-3:2023 addresses biological evaluation and related labeling information.
EN 455-4:2009 remains current for shelf-life determination and includes real-time and accelerated ageing considerations.
A newer EN 455-5:2025 covers measurement of extractable chemical residues used in glove manufacturing.
For EU MDR purposes, EN 455-1:2020+A2:2024 and EN 455-2:2024 were added to the EU list of harmonized medical-device standards in April 2025.
Both Address Pinhole Performance
Barrier integrity is central to both systems.
ASTM D6319 requires nitrile gloves to meet freedom-from-holes requirements, while EN 455-1 specifically focuses on hole detection and sampling.
For bulk purchasing, ask for:
- Test standard
- AQL
- Sampling method
- Lot identification
- Actual test result
A product description saying only “pinhole tested” provides much less information.
Physical Properties Are Also Covered Differently
Both systems evaluate whether the glove can withstand normal medical use.
ASTM D6319 specifies physical requirements for nitrile gloves before and after ageing.
EN 455-2 applies to medical gloves more broadly and specifies physical-property requirements for examination/procedure and surgical gloves.
This means procurement teams should not transfer one test value directly into another standard without checking the relevant method.
EN 455 Adds a Broader Medical-Glove Framework
Because EN 455 is a multi-part series, it extends beyond the core mechanical specification.
The biological-evaluation part addresses information relevant to substances associated with glove use, while EN 455-4 deals with shelf life and degradation.
For long-distance export orders, these areas matter.
A glove can have good initial tensile strength but still require evidence that performance remains acceptable throughout the labeled shelf life.
Does ASTM D6319 Mean FDA Approved?
No.
This is an important procurement distinction.
A glove may be tested according to ASTM D6319, but US medical-market compliance involves FDA requirements beyond the test standard.
FDA states that medical gloves require 510(k) premarket notification.
Before a US private-label order, therefore, verify the exact manufacturer, device, premarket submission, labeling, establishment information, and applicable test documentation rather than accepting “ASTM certified” as the complete answer.
Does EN 455 Mean CE Marked?
Not by itself.
EN 455 provides relevant medical-glove standards, but CE marking depends on conformity with applicable EU legislation and the regulatory route for the finished device.
The harmonized-standard framework can provide a presumption of conformity for the requirements covered by those standards, but it does not turn an isolated laboratory report into complete MDR compliance.
What Should an International RFQ Say?
